Description:

Patients: This Phase II trial involved a total of 350 patients with advanced refractory colorectal cancer. This report focused on the initially planned cohort of 100 assessable patients. There were 52 men and 48 women and the ages ranged from 30-92 (median age was 61).

Treatment: The treatment consisted of chemotherapy (fluoruracil) and biological therapy (bevacizumab – “avastin” a humanized monoclonal antibody targeting vascular endothelial growth factor) and leucovorin.

Toxicity: Four deaths were due to non-progressive disease related causes including pneumonia (3) and unknown etiology (1). In addition grade 3-4 toxicities included hematologic, fatigue, diarrhea, infection, hemorrhage, and cardiovascular.

Results: The median overall survival was 9.0 months. Interestingly, the authors reported a 4% objective response rate, but on independent review (by non-authors) the response rate was calculated to be 1%.
